Properties

It is practically insoluble in water, slightly soluble in methanol, and readily soluble in dichloromethane and chloroform.

ApplicationsN-(Triphenylmethyl)-5-(4'-bromomethylbiphenyl-2-yl)tetrazole (TTBB/BBTT) is a key side-chain intermediate for the synthesis of sartan-class antihypertensive drugs. It is primarily used in the production of olmesartan and irbesartan, while also serving as a general structural unit for the preparation of various angiotensin II receptor blockers (ARBs) such as valsartan and losartan. Specifically, in the synthesis of olmesartan, it undergoes a "one-pot process" with ethyl 2-propyl-4-(1-hydroxy-1-methylethyl)imidazole-5-carboxylate to form the precursor, which hydrolyzes into the active ingredient after oral administration to exert an antihypertensive effect. Additionally, it functions as valsartan Impurity 19 for pharmaceutical quality control and impurity analysis, providing support for the optimization of sartan drug production processes. During the R&D phase of novel angiotensin II receptor antagonists, it serves as a crucial building block for bioactive heterocyclic compounds in laboratory synthesis and small-batch validation, leveraging its pharmacophore structure containing tetrazole and bromomethylbiphenyl groups.
